چکیده

Results Our data of CSF Ab42 and tau levels in conjunction with ε4 allele had shown specificity and sensitivity of 100% and 42.8% respectively for the detection of AD. Ab42 and Apo E ε4 combination had shown specificity 80.8% and sensitivity 72.1 %. The ε4 allele distribution frequency was 40% and 2.5% in AD and NAD respectively, where as ε4/4 genotype and ε3/4 genotype distribution was 10 % and 50 % respectively. Our data has shown that ε4 allele in combination with Ab42 to have better sensitivity and specificity in the diagnosis of AD. AD patients with at least one ε4 allele had significantly lower CSF Ab42 levels than those without ε4 allele (P < 0.001). There was a positive correlation of Ab42 with low MMSE scores. Conclusions Observation from our study suggest that decreased Ab42 and increased tau level in CSF along with Apo E ε4 allele as risk factors for AD. Our study also shows ε4 allele incidence to be a risk factor for AD.

Poor sleep is associated with CSF biomarkers of amyloid pathology in cognitively normal adults.

OBJECTIVE To determine the relationship between sleep quality and CSF markers of Alzheimer disease (AD) pathology in late midlife. METHODS We investigated the relationship between sleep quality and CSF AD biomarkers in a cohort enriched for parental history of sporadic AD, the Wisconsin Registry for Alzheimer's Prevention. A total of 101 participants (mean age 62.9 ± 6.2 years, 65.3% female) ...

Cerebrospinal fluid biomarkers measured by Elecsys assays compared to amyloid imaging

INTRODUCTION Levels of amyloid β peptide 42 (Aβ42), total tau, and phosphorylated tau-181 are well-established cerebrospinal fluid (CSF) biomarkers of Alzheimer's disease, but variability in manual plate-based assays has limited their use. We examined the relationship between CSF biomarkers, as measured by a novel automated immunoassay platform, and amyloid positron emission tomography. METHO...

Apolipoprotein E genotype and the diagnostic accuracy of cerebrospinal fluid biomarkers for Alzheimer disease.

IMPORTANCE Several studies suggest that the apolipoprotein E (APOE) ε4 allele modulates cerebrospinal fluid (CSF) levels of β-amyloid 42 (Aβ42). Whether this effect is secondary to the association of the APOE ε4 allele with cortical Aβ deposition or whether APOE ε4 directly influences CSF levels of Aβ42 independently of Aβ pathology remains unknown. OBJECTIVE To evaluate whether the APOE geno...

Cerebrospinal Fluid Aβ40 Improves the Interpretation of Aβ42 Concentration for Diagnosing Alzheimer’s Disease

The combination of decreased amyloid β42 (Aβ42) and increased total tau proteins (T-Tau) and phosphorylated tau (P-Tau) in cerebrospinal fluid (CSF) has recently been considered as a biological diagnostic criterion of Alzheimer's disease (AD). Previous studies showed significant heterogeneity in CSF Aβ42 levels to discriminate AD from non-AD patients. It was also suggested that the CSF amyloid ...

P 62: Markers of Neuroinflammation Related to Alzheimer\'s Disease Pathology in the Elderly

Alzheimer Disease (AD) is a neurodegenerative disorder and the most common form of dementia. Increasing evidence suggests that Alzheimer's disease pathogenesis is not restricted to the neuronal compartment, but includes strong interactions with immunological mechanisms in the brain. In vitro and animal studies have linked neuroinflammation to Alzheimer's disease (AD) pathology. Studies on marke...
